A product of the Wolves youth academy, Clarke worked his way into the Wanderers first team after a loan spell with Kidderminster Harriers. After dropping out of favour he joined Queens Park Rangers on loan, followed by a two-month spell with Argyle, during which he failed to score and did not impress. In January 2007 he left Wolves to begin a three-year stay at Sheffield Wednesday, where he scored 18 goals in 83 appearances and was also loaned out to Oldham Athletic and Southend United.
After leaving Hillsborough, Clarke's nomadic existence continued as clubs took a punt on his goalscoring potential. In May 2010 he rejoined QPR, followed by Preston North End (loan), Swindon Town, Chesterfield (loan), Charlton Athletic, Crawley Town (loan) and Scunthorpe United (loan) before joining Coventry City on a free transfer in January 2013.
After scoring 28 goals in 45 games for the Sky Blues, Clarke returned to Wolves for a fee reported to be in the region of £750,000 in January 2014, on an 18-month deal with the option of a further year. While with Wolves, he was loaned to Wigan Athletic in February 2015 for the remainder of that season. Clarke's tenure at Wolves came to an end when the option on his contract was not taken up by the club.
He then signed for Bury in June 2015 on a three-year contract just as they had returned to League One. In an impressive 2015-16 season, he scored 18 goals in 37 appearances to become Bury's top scorer. His goal scoring performance made him a target for other League One clubs and in July 2016 he joined Sheffield United on a three-year deal for an undisclosed fee.
